The aim of this investigation was to develop natural superdisintegrant by modify physically properties of patent guar gum using hot air drying technique. Batches of modified guar gum were prepared by subjecting patent guar gum to sequential processes of aqueous wetting and drying using hot air oven. The effect of ratio of guar gum to water and different temperature during formulation of orodispersible tablet of a model drug olanzapine by 32 factorial design was used on disintegration time, wetting time and water absorption ratio of the formulations. The guar gum and modified guar gum were characterized by infra red spectroscopy and scanning electron microscopy and evaluated for physical properties. The least disintegration time of 27sec was obtained using by the ratio of guar gum to water X1 (1:1.25) and temperature X2 (60°). Short term stability studies indicated no significant changes in hardness, friability, in-vitro disintegration time, wetting time, water absorption ratio, drug content and in-vitro drug release.
